Cholera Toxin, produced by the bacterium Vibrio cholerae, is a potent enterotoxin that causes the disease cholera. It is an AB5 toxin composed of one A subunit and five B subunits, which allows it to bind to host cells and enter them, leading to the activation of adenylate cyclase and increased levels of cyclic AMP (cAMP). This results in the excessive secretion of water and electrolytes, causing severe diarrhea and dehydration.

Used in Research Applications:
Cholera Toxin is used as a positive control in cAMP assays for enterotoxins, helping researchers evaluate the activity of other potential enterotoxins and their effects on cellular cAMP levels.
Used in Cell Culture Media:
Cholera Toxin is added as a supplement in cell culture media for primary tumors and epithelial cells. This application aids in studying the effects of the toxin on cell growth, differentiation, and signaling pathways, as well as its potential role in cancer progression and epithelial cell function.

Biochem/physiol Actions

Toxin consisting of an A subunit (27 kDa) surrounded by five B subunits (approximately 12 kDa each), which attach the toxin to ganglioside GM1 on the cell surface. The A subunit catalyzes ADP-ribosylation of the α-subunit of the stimulatory G protein (Gαs), reducing GTPase activity and activating the α-subunit. This activation of Gαs leads to an increase in the activity of adenylyl cyclase, resulting in increased levels of cAMP. Also ADP-ribosylates transducin in the eye rod outer segments, inactivating its GTPase activity. Cholera toxin has also been reported to ADP-ribosylate tubulin. Shown to be a potent mucosal vaccine adjuvant, inducing T helper cell type 2 responses by inhibiting the production of interleukin-12.
