ML167 is a highly selective cell permeable inhibitor of Cdc2-like kinase 4 (Clk4) with an IC50 of 136nM. It exhibits at least 10-fold greater selectivity over related kinases such as Clk1, Clk2, Clk3, Dyrk1A, and Dyrk1B.

Disclosed herein are novel quinazolin-4-amine derivatives that are inhibitors of Clk1, Clk2, Clk3, Clk4, or Dyrk1A. Also disclosed are quinazolin-4-amine derivatives as potent and selective inhibitors of Clk1, Clk4, and Dyrk1A.
